Here, we synthesize and characterize a photoreactive GTP affinity probe that covalently photocross-links to protein targets and has an alkyne handle for click chemistry conjugation to reporter tags. Affinity proteomics seeks to use them to analyse proteomes using collections of specific binding reagents. While the most familiar affinity reagents are the classical antibodies and their fragments, there are also alternative engineered forms of novel protein scaffolds and nucleic acid aptamers with similar diversity and binding properties. Sacco et al. found that the affinity proteomics data identified a number of protein interactors that co-enriched with the phosphatase enzymes under examination, with around 10% of the data reported elsewhere in research literature.
